In answer to your question, the hole in the release lever that is closest to the shaft will give you the most travel. That said, use the middle hole. This isn't your problem. Your first problem was that it wouldn't shift into second. Gear. Presumably it was shifting in and out of all the other gears, in which case, you don't have a hydraulic problem. 


You say it worsened to the point where it wouldn't go into any gear. What was the clutch pedal feeling like at this point? Had its behavior changed since you left on your trip to CVS? 


If the master cylinder is as old as the slave, and you lost 1/4" of fluid from the clutch reservoir over the course of a short drive, inspect for fluid leaks from the master. It may be bypassing. Look inside the car where the cylinders come through the firewall. This is where you'll usually see the first signs of a leak. If the master cylinder appears dry, you're lucky. Is the clutch the same? 1/4" of fluid went somewhere though there's a slim chance you just had some air left over from your bleeding that has sorted itself out with some use, and sucked in the fluid. 


Another lister mentioned the release lever/shaft. The fork that attaches to that shaft, which holds the throwout bearing (or "release bearing"), is held onto the release shaft with a screw that is known to break. Savvy knuckleheads drill the fork and shaft and put an extra pin in, when replacing they have the tranny out. Safely jack up the car, disconnect the slave push rod from the release lever and use whatever's handy (pipe is nice) to move the release lever by hand. As you push it backwards, it will travel freely for about 1/4 inch. It shouldn't travel more than that. More on this in a minute. You should feel the lever resist your pressure. The throwout bearing is now against the clutch pressure plate. Keep pushing the lever backward and you should feel constant pressure as it travels to the end of its range, This pressure is that of the clutch pressure plate diaphragm (spring). If you can move the lever backward without much resistance or if it feels jerky and rough, it's likely that the screw has busted and the fork is moving on the shaft. This can be weird because sometimes the fork shifts on the lever more than at other times, when the screw breaks. If it is broken, it will feel just plain wrong. You'll know. 


If there is excessive travel of the lever when you first start pushing on it, before you feel the throw out bearing contact the pressure plate, check the master and slave cylinder push rods. If the holes where they pin to the clutch pedal and the release lever respectively are worn and oval-shaped, it is possible that the hydraulics and release shaft/fork pin are fine but the release fork just isn't traveling far enough to completely release the pressure plate when you tromp on the clutch pedal. This has happened to me. If you used the new push rod that came with the slave cylinder, you should be fine but check the master cylinder push rod. 


The other possibility, and this has happened to me, twice (!!!), is that the pressure plate itself has failed. This usually doesn't happen suddenly, though, so we'll leave that genie in the bottle until you check out the other stuff.

Dear fellow knuckle heads ( for owning these demon Triumphs) :)) 
I changed the slave cylinder (clutch), bled it, bla, blah,bla . Due to my overallhealth, the room between the car and my garage wall, and mental health?it took me quite a while ( months ) to finishby any competent mechanic, the two hour job.Purchased my '74 TR6 in 1998. Ok, so much for the back round info. ?I took it for a short drivejust ?to test my work, it worked well. It sat two weeks before I bravely set out on a 2 mile roundtrip to CVS. ?Turning off the main road into the parking lot and !!! SURPRISE !!!! it wouldn't go into 2nd gear.It wasn't pretty driving home but got it into the yard. ?Wouldn't go into any gear.The only thing I could think of besides toping off the clutch fluid, down 1/4", Would possiblybe the clutch lever which has three holes in it. ?I don't remember which hole?I used. ?Assuming I had it in the center hole, which hole would move the clutch most (and hopefully)make the damn thing work. ?I'm tired of being afraid to drive it and have absolutely NO MORE MONE 
Y to spend on repairs.?Probably spent $3,000 in the last year and a half. ??

This might be a red herring but on several occasions I have had the clutch disk stick to the flywheel. Usually after extended storage. This requires a bit of abuse to get it unstuck. Brian Alwin 72 TR6

You either have a bad clutch slave or master cylinder or the clutch fork pin has broken. If pumping the clutch repeatedly makes it better then hydraulic. You can look under the car (on a lift or well placed jack stands) and obseven the range of motion of the slave rod and lever it is attached to. It should have a little play, but then keep moving through the whole clutch pedal depression. If it doesn't once again hydraulics. If it does appear to move all the way through the pedal depression it could be a broken pin.

Have you made sure that the breather hole in the cap of the clutch fluid 
reservoir is open? It sometimes plugs with dried fluid + crud. My '72 is 
long gone, but I love sharing ongoing misery.
